ピラセタムとのスタックとして人気なCDPコリン・アルファGPCは、「朝に摂取すべき」という意見が多くみられれます。その主張のエビデンスをまとめてみました。
半減期の長さ
健康的なインド人を対象に、2社の1000mg(CDPコリン)を1回の経口摂取で調べた結果、半減期は66.348+/-8.445時間で、排出に数日要するとしています[1]。また、同調査では、CDPコリンの吸収率は97.55-100%であると報告しています。
ヒトにおける、用量による違いを調べた別の調査では、CDPコリン経口摂取により、
- コリンは2-3hがピークで、10h持続する
- ウリジンは、90分時点で、500mg(101%)、2000mg(136%)、4000mg(134%)の上昇を見せ、6時間持続する
以上の2つの調査から、ピークは2-3時間だが、排出率が悪く半減期が数日である。故に、推奨されている250-500mg/日を継続で蓄積する可能性がある。
この結果、疲労感・頭痛などを引き起こすと考えられる。
記憶の定着
記憶の定着には、脳が活性化されていない状態で、低いアセチルコリン濃度が必須であるとみられている[4]。しかし、多くの動物実験では、健常ラットに対して、コリン塩やCDPコリン、代謝物であるウリジン投与で、脳内アセチルコリン濃度の上昇がみられている[2][3]。
そのため、寝る2-3時間前にCDPコリンの摂取は避けるべきだろう。科学的に検証されていないが、これが理由に、アセチルコリンを枯渇させるピラセタムを夜に摂取する人もいる(エビデンスは全くないが・・・)。
- 健常者は少量が(50-100mg)ベストっぽい
- ピラセタムと比率にこだわらない
- 半減期が長いから朝一摂取が望ましい

The half-life of elimination for the choline spike seen with supplemental CDP-choline (1000mg) appears to be fairly delayed, at 66.348+/-8.445 hours.
Among aged animals consuming a UMP-containing diet (2.5%, w/w) for 1 or 6 weeks, baseline ACh levels in striatal dialysates rose from 73 fmol/min to 148 or 197 fmol/min . Consuming a lower dose (0.5%) for 1 week produced a smaller but still significant increase (from 75 to 92 fmol/min,, and elevated striatal ACh content (by 16%;. Dietary UMP (0.5%, 1 week) also amplified the increase in ACh caused by giving atropine (10 microM in the aCSF); atropine alone increased ACh concentrations from 81 to 386 fmol/min in control rats and from 137 to 680 fmol/min in those consuming UMP . Young rats eating the UMP-containing diet exhibited similar increases in basal ECF ACh (from 105 to 118 fmol/min) and in the increase produced by atropine (from 489 to 560 fmol/min;. These data suggest that giving a uridine source may enhance some cholinergic functions, perhaps by increasing brain phosphatide levels.

New findings by Gais and Born (10) presented in this issue of PNAS provide compelling evidence in human subjects that SWS and the accompanying low levels of acetylcholine during SWS may mediate a critical memory consolidation process. These findings support two-stage models of memory consolidation, as will be discussed below.
